Manning, who missed all of the 2011 season after undergoing neck surgery, completed 19-of-26 passes for the Broncos (1-0).
Demaryius Thomas caught five passes for 111 yards and a touchdown, while Willis McGahee carried the ball 16 times for 63 yards in the victory.
Ben Roethlisberger went 22-for-39 through the air for 245 yards, two touchdowns and an interception for Pittsburgh (0-1).
Antonio Brown paced all Steelers receivers with 74 yards on four catches.
